Phase-sensitive Optical Coherence Microscopy Measurements in Living Cells

Phase-sensitive optical coherence microscopy (OCM) is a quantitative phase imaging technique capable of measuring sub-wavelength optical path delay differences between two light beams. The method is extremely sensitive to phase changes, with optical path delay measurement accuracy of several tens of picometers, enabling accurate optical thickness change measurements in live cells in a label-free manner.

We developed and constructed the spectral-domain phase microscopy (SDPM) system, a novel and ultrahigh accuracy fiber-optics system that is based on the OCM principle. Using the system, it is possible to obtain several tens of picometer accuracy when measuring changes in optical thickness of live cells without any labeling, without sample contact, and without special sample preparation. We demonstrate the system usefulness on three different applications: (a) measuring changes in the visco-elastic properties of red blood cells where their membrane fluctuations are limited, (b) measuring hyperthermia effect on the red blood cells visco-elsasticity, and (c) optical measurements of neuronal activity in mammalian axon.  The proposed system is compact, portable, inexpensive, and has great potential for clinical application.
